1. Knowledge of research theories and methods of molecular genetics/genomics (structural and functional), bioinformatics and molecular genetic plant-pathogen interactions.
a. Nucleic acid analysis including northern blot, RNase protection, quantitative RT-PCR, siRNA and miRNA gel blot hybridizations, etc
b. Developed an agrobacterium leaf-infiltration system for gene silencing in papaya as a potential functional genomics tool
c. Identified the P0 protein of Sugarcane yellow leaf virus to be a suppressor of RNA silencing, and characterized the activity and functional domains by deletion analysis in Nicotiana benthamiana
2. Knowledge of eukaryotic genome organization, gene structure and gene regulation.
a. Isolated and characterized two strong constitutive promoters from sugarcane,
b. Characterized stable transgenic rice and sugarcane lines expressing these promoters,
c. US patents issued for these two promoters
d. Produced biologically active human GMCSF in transgenic sugarcane, including field trials
3. Ability to concieve, plan and conduct research using a bionformatics/computational biology
a. Bioinformatic analysis to identify potential miRNA genes in the papaya genome using miRcheck (Jones-Rhoades & Bartel, 2004), a PERL script which uses RNAfold (Vienna RNA package 1.6.3, http://www.tbi.univie.ac.at/~ivo/RNA/) to predict secondary structure of the nt sequence,
b. Bioinformatic analysis to predict tRNA genes in the papaya genome using tRNAscan-SE (Lowe & Eddy, 1997),
4. Skill in presenting information to scientists, producers, and government agencies using conventional, electronic, or multi-media technologies.
